Introduction
Zap-Hybridoma™ is a cell culture media supplement specifically designed to improve the production of monoclonal antibodies in hybridoma and myeloma cells. It is IgG free and chemically defined, and delivers growth and productivity similar to fetal bovine serum (FBS). Zap-Hybridoma is produced in an animal-free production system and delivers lot-to-lot consistency. Zap-Hybridoma contains animal-free recombinant protein and other factors essential for growth.
When using Zap-Hybridoma, FBS supplementation can be reduced or eliminated, and it allows the direct transfer from FBS containing media to serum-free media with little or no adaptation. It can also replace other media supplements such as insulin-transferrin cocktails, and supplements that contain animal-derived components. Zap-Hybridoma has many effects in cell culture including enhanced cell growth, reduced doubling times, increased maximum cell density, reduced apoptosis, and increased viability.
Applications
Zap-Hybridoma can be used during antibody production, cell line maintenance, fast adaptation of cells to reduced-serum or serum-free media, and single cell cloning. Zap-Hybridoma is recommended for use in hybridoma cell culture when using classical, serum-free, animal-free, or chemically defined media. Zap-Hybridoma is IgG free and its components can be easily separated from antibodies during downstream purification.
Instructions for use
Zap-Hybridoma is provided as a 100x sterile concentrate, 10 mL of Zap-Hybridoma will supplement 1 L of media. Some cell types benefit from supplementing media at 2x final concentration. At 2x final concentration 10 mL of Zap-Hybridoma will supplement 0.5 L of media.
For best results using classical formulations, InVitria recommends using DMEM/F12 or IMDM media supplemented with Zap-Hybridoma. Typically no insulin or transferrin is required when using Zap-Hybridoma.
