Introduction

InVitria’s Zap-SR supplement is designed to reduce serum in cell culture medium while maintaining cell proliferation and high cell viability. Zap-SR is compatible with F12, DMEM/F12, DMEM, IMDM, and other basal medium formulations. The ingredients of Zap-SR do not contain any animal-derived components and is prepared as a sterile-filtered 100x concentrate.

Instructions for Use

Requirements for extracellular matrix, growth factors, and other serum-derived components are highly specific for individual cell lines. Therefore, we recommend an initial serum reduction study per desired Zap-SR application as specified below in Table 1. Cells should be passaged directly from the 10% serum-containing medium into the reduced serum medium with and without Zap-SR and standard subculture times and protocols should be followed. The highest concentration of serum that fails to support adequate cell growth without Zap-SR but is rescued by the addition of Zap-SR is the optimum FBS/Zap-SR mixture. Further reductions in serum can be performed as desired.

Final serum concentrations will vary depending on the cell type. In order to identify the optimal serum and Zap-SR concentrations, 25 mL of the 7 medium formulations should be composed. The highest concentration of serum that fails to support adequate cell growth without Zap-SR but is rescued by the addition of Zap-SR is the optimum FBS/Zap-SR mixture.
